« Back to Glossary IndexAgomelatine (Valdoxan) is an atypical, non-SSRI antidepressant that functions primarily as a melatonin receptor agonist and a serotonin antagonist. It treats major depressive disorder (MDD) by restoring circadian rhythms, promoting slow-wave sleep, and increasing dopamine/noradrenaline release in the frontal cortex. Structurally similar to melatonin, it binds to receptors in the suprachiasmatic nucleus (SCN)), advancing circadian rhythms to normalize sleep-wake cycles. By blocking serotonin receptors, it increases the release of dopamine and noradrenaline in the prefrontal cortex, aiding mood improvement. Unlike traditional antidepressants, it typically causes no daytime side effects.
